Manufacturing a custom mRNA vaccine tailored to an individual patient tumor mutational profile within weeks requires an unprecedented bio-manufacturing pipeline. Moderna process begins with automated next-generation sequencing (NGS) of patient tumor biopsies and healthy blood samples.

Bioinformatic algorithms compare the genomic sequences to identify unique somatic mutations. An AI-trained epitope prediction model evaluates HLA binding affinity to select up to 34 neoantigens most likely to trigger a robust T-cell response. The chosen sequences are automatically synthesized into a single mRNA transcript packaged in lipid nanoparticles (LNPs).
